Has anyone had any experience using a .45 colt revolver with six inch barrel on deer size game. I realize it would need to be fairly close.

you don't have to be as close as you think, you just have to change your shooting style. practice shooting from a seated position using your knees as braces, or at the very least use sticks or a tree. I shot my 454 into a 5 gallon bucket offhand at about 75 yards pretty regularly. It just takes practice, and a longer barrel would help. I'd say 50-75 yards would be no problem. Pick a bullet stay with it and practice practice practice.
